A novel method to establish the rabbit model of knee osteoarthritis: intra-articular injection of SDF-1 induces OA

Abstract Background Animal model of Knee Osteoarthritis (OA) is the primary testing methodology for studies on pathogenic mechanisms and therapies human OA disease. Recent major modeling methods are divided into artificially induced spontaneous. However, these have some disadvantages slow progression, high cost no correlation with pathogenesis OA. Methods Our attempted to find a rapid, easy, consistent natural pathological process method by intra-articular injection stromal cell-derived factor 1 (SDF-1) in rabbit knee. After induction we collected cartilage specimens from medial femoral condyle undergo macroscopic, histological, immunohistochemical, biochemical evaluations. Meanwhile, compared Hulth surgical evaluate its efficacy. Results Macroscopic observation modified Mankin score histological staining exhibited typical features middle stage SDF-1 injected groups. Immunohistochemically, positive expression interleukin-1 (IL-1) tumor necrosis α(TNF-α) was earlier higher dose group than group. The matrix metalloproteinases (MMPs) synovial fluid chondrocytes significantly increased, but type II collagen (COLII) aggrecan (ACAN) protein expressions decreased following extension time increase concentration. Conclusions data indicated (40μg/kg, three times 12 weeks) can induce knee successfully more rapidly easily traditional modeling. study provided further option establishment animal model.
